Preparation of 5'monophosphate mononucleotides from single stranded DNA

• 1st day:
• 2...5 µg ssDNA (e.g. M13)
• add 10 µl 10 x lysis buffer (300 mM NaOAc pH 5.2;  1 mM ZnCl₂)
• adjust to 99 µl and add 1 µl Nuclease P1 (1 g/l)
• incubate ON at 37°C
 
• 2nd day:
• add another 1 µl Nuclease P1
• incubate at 72°C for 1 hour
